党参多糖对脑缺血再灌注损伤大鼠血清 S100-β 及 NSE 水平的影响

Effects of Codonopsis Pilosula Polysaccharide on Serum S100-β and NSE Levels in Rats with Cerebral Ischemia-Reperfusion Injury

摘要: 目的 观察党参多糖对缺血再灌注损伤大鼠血清 S100-β 蛋白( S100-β) 及神经特异性烯醇化酶( NSE) 含量的影响。 方法 随机将 SD 大鼠分成 5 组:假手术组、模型组、依达拉奉组和 2 个党参多糖高、低剂量组,每组 10只。 采用线栓法阻断大鼠中脑闭塞(MCAO)2 h 后再灌注 24 h 建立脑缺血再灌注模型,2 个党参多糖和依达拉奉组大鼠术前连续 14 d 给予相对应的药物,于再灌注后 24 h 取样,采用尼氏染色和 HE 染色法进行组织学观察,测量脑含水量及伊文思蓝法以评估卒中,ELISA 法分别检测各组血清中 NSE 和 S100-β 水平。 结果 与假手术组比较,模型组大鼠脑组织含水量及通透性明显增加(P<0. 01) ,血清中 NSE 及 S100-β 蛋白含量明显增加(P<0. 01) 。 与模型组相比较,党参多糖高、低 2 个剂量组血清中 NSE 和 S100-β 蛋白含量明显降低( P<0. 01) ,且党参多糖高剂量组脑组织含水量及通透性明显降低(P<0. 01) 。 结论 党参多糖能明显减轻大鼠脑缺血再灌注损伤的程度,降低血清中 NSE 和 S100-β 水平,具有良好的神经保护作用。

Abstract: Objective To observe Codonopsis pilosula polysaccharide ( CPPS) on ischemia-reperfusion injury of rat serum S100-β protein and neuron-specific enolase ( NSE) levels. Method SD rats were randomly divided into 5 groups, namely sham operation group, model group, Edaravone group and 2 codonopsis pilosula polysaccharide (CPPS) high-dose and low-dose groups, with 10 rats in each group. By using line switch block rat middle cerebral artery occlusion ( MCAO) 2 h reperfusion 24 h after cerebral ischemia reperfusion model is set up, two CPPS and edaravone group rats were given corresponding drugs for 14 consecutive days before surgery, Nissl staining and HE were used for histological observation 24 h after reperfusion. Cerebral water content was measured and Evans blue method was used to assess cerebral apoplexy. Serum NSE and S100-β levels were determined by ELISA. Result Compared with sham operation group, the water content and permeability of brain tissue in model group were significantly increased ( P < 0. 01) , and the contents of NSE and S100-β protein in serum were significantly increased ( P < 0. 01 ) . Compared with model group, serum NSE and S100-β protein contents in CPPS high-dose and low-dose groups were significantly decreased ( P < 0. 01) , and brain tissue water content and permeability in CPPS high-dose group were significantly decreased ( P < 0. 01) . Conclusion CPPS can significantly reduce the level of NSE and S100-β in serum of rats with cerebral ischemia-reperfusion injury, and have good neuroprotective effect.
